However, in practice, these components are often added separately into the reaction wells, which can disrupt the balanced competition and lead to deviations from the expected proportional relationship. This issue becomes more pronounced under conditions of extended incubation times or elevated temperatures, significantly affecting the accuracy of the results. Changhai Hospital at the Second Military Medical University conducted a study to evaluate this effect. In their experiment, 94 negative specimens were diluted 1:30 with physiological saline. After the specimens were added to the wells, the enzyme-labeled antibodies were introduced at different time intervals. However, when the enzyme-labeled antibody was added after a two-minute delay, a 7.4% false positive rate occurred. After 30 minutes, the false positive rate increased dramatically to 57.4%. These findings strongly suggest that for optimal results, the enzyme-labeled antibody should be added immediately after the sample to maintain the integrity of the competitive binding process.
